Anotation
Liečebný potenciál mezenchýmových kmeňových buniek (MSC) je často sprostredkovaný ich parakrínnou funkciou. Analýza sekrečných faktorov a možnosť ich využitia v regeneračnej medicíne je v súčasnej dobe intenzívne študovaná s cieľom zavedenia tohto typu liečby do klinickej aplikácie. Kondiciované médium (KM) a exozómy (EV) uvoľnené bunkami obsahujú dôležité bioaktívne molekuly (proteíny, lipidy, metabolity, miRNA, mRNA, ncRNA, DNA), ktoré majú svoj špecifický obsah. Exozómy sú schopné dodávať alebo prenášať bio-informácie a relevantné genetické informácie a proteíny medzi blízkymi alebo vzdialenými bunkami a môžu následne modulovať recipientné bunky. Cieľom projektu je zistiť genomický a proteomický profil KM a EV izolovaných z psích MSC adultného a neonatálneho pôvodu. Zároveň v in vitro podmienkach overiť ich biologickú aktivitu a selektovať tie KM alebo EV, ktoré potenciujú endogénnu regeneráciu a ovplyvňujú imunitnú reakciu. Súčasťou poslednej etapy projektu bude štúdium a vyhodnotenie účinnosti nanoterapie sprostredkovanej KM a EV u psov s vybranými neurodegeneračnými ochoreniami (kanínna kognitívna dysfunkcia, degeneratívne zmeny sietnice, traumy rohovky, poranenie miechy), ktorá môže byť vhodnou náhradou bunkovej terapie vo veterinárnej praxi a ideálnym animálnym modelom pre humánnu medicínu.
The therapeutic potential of mesenchymal stem cells (MSC) is often mediated by their paracrine function. The analysis of secretory factors and the possibility of their use in regenerative medicine is currently being studied intensively with the aim of introducing this type of treatment into clinical application. Conditioned media (KM) and exosomes (EV) released by cells contain important bioactive molecules (proteins, lipids, metabolites, miRNA, mRNA, ncRNA, DNA) that have their specific content. Exosomes are able to deliver or transmit bio-information and relevant genetic information and proteins between nearby or distant cells and can subsequently modulate recipient cells. The aim of the project is to determine genomic and proteomic profile of KM and EV isolated from canine MSC of adult and neonatal origin. At the same time, in vitro conditions should verify their biological activity and select those KMs or EVs that potentiate endogenous regeneration and affect the immune response. Part of the last phase of the project will be the study and evaluation of the efficacy of KM and EV-mediated nanotherapy in dogs with selected neurodegenerative diseases (canine cognitive dysfunction, retinal degenerative changes, corneal trauma, spinal cord injury), which may be a suitable may be a suitable substitute for cellular therapy in veterinary practice and an ideal animal model for human medicine.
