Anotation
Posun paradigmy smerom k parakrinnej signalizácii ako primárnemu mechanizmu terapeutickej účinnosti mezenchýmových kmeňových buniek (MSC) smeruje k využitiu kondiciovaného média (KM) pri neuroregenerácii. KM pozostáva z rozpustných faktorov (sekrétom) a extracelulárnych vezikúl (ECV) uvoľňovaných z MSC s obsahom funkčných bioaktívnych molekúl. Prekondiciovanie MSC v hypoxickom alebo prozápalovom mikroprostredí zlepšuje ich imunomodulačné, antiapoptotické, angiogénne a proregeneračné vlastnosti. Cieľom projektu je v in vitro podmienkach optimalizovať stimulačnú hypoxicko-prozápalovú intervenciu MSC za účelom vytvorenia “KM a ECV novej generácie“ s vyššou biologickou aktivitou, ktorú overíme na neuronálnych bunkových líniách a primárnych kultúrach. Následne vykonáme kombinovanú terapiu, podaním KM (i.v) a ECV (lokálne, do lézie) s najvyšším neuroregeneračným profilom u psov s akútnym poranením miechy. Stimulované KM a ECV môžu predstavovať inovatívnu acelulárnu terapiu nielen pre veterinárnu prax.
A paradigm shift toward paracrine signaling as the primary mechanism of therapeutic efficacy of mesenchymal
stem cells (MSCs) is leading to using conditioned medium (CM) in neuroregeneration. KM comprises soluble
factors (secretomes) and extracellular vesicles (ECVs) released from MSCs containing functional bioactive
molecules. Preconditioning MSCs in a hypoxic or pro-inflammatory microenvironment improves their
immunomodulatory, anti-apoptotic, angiogenic and pro-regenerative properties. The goal of the project is to
optimize the stimulating hypoxic-proinflammatory intervention of MSCs in vitro conditions in order to create \"CM
and ECV of a new generation\" with higher biological activity, which we will verify on neuronal cell lines and
primary cultures. Subsequently, we will perform a combined therapy, administering CM (i.v.) and ECV (locally)
with the highest neuroregenerative profile in dogs with spinal cord injury. Stimulated CM and ECV can represent
an innovative therapy for veterinary practice.
